Current Projects
Evaluation of Immunotoxic Chemicals
The main focus of my lab is to evaluate the immunotoxicity caused by exposure to occupationally relevant chemicals using a standardized sequence of immunotoxicological assays which will evaluate the effects of these chemicals on the immune system. The test articles/chemicals we evaluate are identified through collaborations with members of industry, other divisions of NIOSH and other branch members within the Health Effects Laboratory Division of NIOSH, and the National Institute of Environmental Health Sciences (NIEHS). Through these collaborations, sensitizing components and chemicals present in manufactured goods including: metal working fluids, thermal transfer ribbon and wound closure bandages have been identified using the local lymph node assay (LLNA) (Myers et al. 2007; Anderson et al. 2009a); immunotoxicological evaluations of welding and asphalt fumes have been conducted (Anderson 2008; Anderson 2007) and the adverse health effects after exposure to other high production volume chemicals such as 1-bromopropane (1-BP) have been characterized in the laboratory (Anderson et al. 2009b). In addition, the lab also has experience utilizing a murine model of asthma in the evaluation of chemicals contributing to occupational asthma (Fairley et al. 2007).
Investigations into Health Effects caused by exposure to indoor air reaction products
Other research projects and interests include the identification, quantification and evaluation of immunotoxicity of the reaction products of gas-phase compounds, especially dicarbonyls, present in the indoor environment using a novel in vitro exposure system that utilizes the air/cell interphase with hopes to identify biomarkers that can be used to screen indoor environments. This project is a synergistic approach combining toxicology and indoor chemistry. Past successes include hazard identification of many of these indoor air reaction products as irritants and sensitizers and investigations into changes in gene expression after exposure to the indoor air reaction products.
The Contribution of Chemical Adjuvancy to Occupational Asthma
During the past several decades there has been a remarkable and unexplained increase in the prevalence of asthma. While the hygiene hypothesis provides one potential explanation, individuals in industrial societies are exposed to an increasing number of chemicals. While many chemicals are known to directly induce asthma, there is the potential for chemicals to function as adjuvants, augmenting the immune response induced by other chemical and protein allergens. Recent studies in our lab have demonstrated the potential of several non-allergenic chemicals including Bisphenol A (BPA), triclosan, and perflourooctanonic acid (PFOA), to act as adjuvants, augmenting the IgE and airway hyper-reactivity responses to the protein, ovalbumin. Currently the EPA’s harmonized test guidelines for Immunotoxicity testing do not require a test for adjuvancy and no validated assays are available for this purpose. Therefore, understanding the mechanism(s) by which chemicals enhance allergic-like responses is important to aid in the development of more effective methods to screen for chemical adjuvancy.
